Click Chemistry has found a wide range of applications in bioconjugation in the pharmaceutical and biotechnology industries. It is a term introduced by KB Sharpless in 2001 to describe a high yield, a wide range reaction that only produces by-products. These by-products can be separated without chromatography, are stereospecific, easy to perform, and can be conducted in easily removable or benign solvents. 

Biochempeg provides many
PEG Azide and PEG Alkyne derivatives for the conventional click chemistry reaction with copper (I) (CuAAC), but due to the cytotoxicity of the catalyst, the application of this reaction in biomedical systems is limited.

The novel copper-free click chemistry is based on the reaction of a cyclooctyne (DBCO) moiety with an azide-labeled reaction partner (called strain-promoted alkyne azide cycloaddition (SPAAC)). This new "click chemistry" is very fast at room temperature and does not require cytotoxic Cu (I) catalysts. Cyclooctyne has high thermal stability, very narrow and specific reactivity with azide, resulting in almost quantitative yields of stable triazoles. This unique covalent bond is generated when DBCO incorporated into one biomolecule reacts with the azide linker incorporated into the second biomolecule.


Features and Advantages Of Copper-Free Click Chemistry
