Biochempeg provides many PEG Azide and PEG Alkyne derivatives for the conventional click chemistry reaction with copper (I) (CuAAC), but due to the cytotoxicity of the catalyst, the application of this reaction in biomedical systems is limited.
The novel copper-free click chemistry is based on the reaction of a cyclooctyne (DBCO) moiety with an azide-labeled reaction partner (called strain-promoted alkyne azide cycloaddition (SPAAC)). This new "click chemistry" is very fast at room temperature and does not require cytotoxic Cu (I) catalysts. Cyclooctyne has high thermal stability, very narrow and specific reactivity with azide, resulting in almost quantitative yields of stable triazoles. This unique covalent bond is generated when DBCO incorporated into one biomolecule reacts with the azide linker incorporated into the second biomolecule.
