Biochempeg provides many PEG Azide and PEG Alkyne derivatives for the conventional click chemistry reaction with copper (I) (CuAAC), but due to the cytotoxicity of the catalyst, the application of this reaction in biomedical systems is limited.
The novel copper-free click chemistry is based on the reaction of a cyclooctyne (DBCO) moiety with an azide-labeled reaction partner (called strain-promoted alkyne azide cycloaddition (SPAAC)). This new "click chemistry" is very fast at room temperature and does not require cytotoxic Cu (I) catalysts. Cyclooctyne has high thermal stability, very narrow and specific reactivity with azide, resulting in almost quantitative yields of stable triazoles. This unique covalent bond is generated when DBCO incorporated into one biomolecule reacts with the azide linker incorporated into the second biomolecule.
Features and Advantages Of Copper-Free Click Chemistry
• Biocompatibility – no cytotoxic copper catalyst is required – good in vivo application.
• Mild conditions –conjugation in an aqueous buffer medium and at low temperatures
• Stability – DBCO and azide moieties are stable for a long time
• Efficiency – stable triazole formation in quantitative yield
• Specificity and biological orthogonality – Azides only react with DBCO in the presence of -NH2, -SH, -COOH and other protein functionalities
However, most cyclooctyne reagents do not have good water solubility, which limits their application in biomedicine. Compared with cyclooctyne, Biochempeg's DBCO PEG derivatives have good water solubility, and have better biocompatibility and no cytotoxicity than traditional click reagents. Biochempeg has enhanced its ability to provide high-quality PEG DBCO products in large kilograms and above at very affordable prices.
The following are a few examples of PEG DBCO products produced by Biochempeg. PEG and monodisperse PEG DBCO derivatives with different branches, molecular weight and functionality can be obtained by custom synthesis.
