Polyethylene glycol (PEG)-lipid conjugates are PEG derivatives containing lipid molecules such as DMG or DSPE, which have been used in bioconjugation, and lipid nanoparticle (LNP) drug delivery to improve circulation times for liposome-encapsulated drugs. The lipid molecule has the characteristics of low toxicity and easy preparation, and it can not only be used as a carrier for water-soluble drugs, but also as a carrier for fat-soluble drugs.
PEG lipids can significantly influence the properties of lipid nanoparticles in various ways:
1. Particle Size: The amount of PEG lipids can affect the size of the nanoparticles.
2. Particle Stability: By decreasing particle aggregation, PEG lipids enhance the stability of the nanoparticles.
3. Blood Circulation Time: Optimizing PEG can prolong the blood circulation time of nanoparticles by reducing clearance by the kidneys and the mononuclear phagocyte system.
4. Targeted Delivery: PEG lipids can also be used to attach specific ligands to the nanoparticles, allowing for targeted delivery.
