Maleimide-PEG-Hydroxyl (MAL-PEG-OH) is a linear heterobifunctional PEG reagent with a maleimide and a hydroxyl group.  With a maleimide group at one end, MAL-PEG-OH enables the conjugation of thiol-containing molecules or surfaces through Michael addition reactions, forming stable thioether linkages. Meanwhile, the hydroxyl group at the other end provides versatility for various conjugation strategies, such as reaction with carboxylic acids to form ester bonds.

MAL-PEG-OH finds utility as a crosslinking reagent due to its PEG spacer, which enhances water solubility and biocompatibility. Additionally, it serves as a precursor for the synthesis of PEGylated block copolymers, such as PLA-PEG and PCL-PEG, via hydroxyl-initiated polymerization with lactide or caprolactone.
